Methodology_Description:
Genomic DNA was extracted from Snowy Plover blood and tissue samples as described in Haig et al. (1997). The polymerase chain reaction (PCR) was used to amplify a 322 bp fragment of the mitochondrial control region, or d-loop, using the primer pair TS437L and TS778H (Wenink et al. 1994). Each reaction was carried out using 100 ng of genomic DNA in a 50 µL volume. Fragments were amplified using a PTC 100 thermal cycler (MJ Research). An initial denaturation of 10 min at 93°C was followed by 44 cycles of 1 min denaturation at 92°C, 1 min annealing at 45°C, and 2 min extension at 72°C, with a single final extension at 72°C for 10 min. The reactions were purified electrophoretically; bands of approximately 350 bp were extracted from a 1% agarose gel using an ultra-free-mc 0.45 filter (Millipore) and purified over a micron-50 filter (Millipore). Sequence data were generated with an automated sequencer (Applied Biosystems model 373A) located in the Central Services Laboratory at Oregon State University.
